MG63Á¶°ñ¼¼Æ÷¸¦ ´ÏÄÚƾ ³óµµ°¡ °¢°¢ 1.0 ¥ìM, 1.0 mM, 2.5 mM, 5.0 mM, 7.5 mM, ±×¸®°í 10.0 mM,ÀÌ ÇÔÀ¯µÈ ¹èÁö¿¡¼­ 1ÀÏ, 2ÀÏ, 3ÀÏ ±×¸®°í 6Àϵ¿¾È ¹è¾ç ½ÇÇèÀ» ÇÏ¿© ´ÙÀ½°ú °°Àº °á°ú¸¦ ¾ò¾ú´Ù.
MG63Á¶°ñ¼¼Æ÷ÀÇ Áõ½ÄÀÌ Àú³óµµÀÇ ´ÏÄÚƾ¿¡¼­´Â ÀϽÃÀûÀ¸·Î È°¼ºÈ­µÇ¾úÁö¸¸ 5.0 mM ÀÌ»óÀÇ °í³óµµ¿¡¼­´Â ¾ïÁ¦µÇ¾ú´Ù. ¿°±â¼º ÀλêºÐÇØÈ¿¼ÒÀÇ È°¼ºÀº ´ÏÄÚƾ ³óµµ°¡ Áõ°¡ÇÔ¿¡ µû¶ó °¨¼ÒÇÏ¿´´Ù.
¿À½ºÅ׿ÀÄ®½Å »ý¼º¾çÀº, ¹è¾ç 1ÀÏ ÈÄ, 5.0mM ÀÌÇÏÀÇ ³óµµ¿¡¼­ Áõ°¡ÇÏ¿´À¸³ª 7.5 mM ÀÌ»óÀÇ °í³óµµ¿¡¼­´Â °¨¼ÒÇÏ¿´´Ù. MG63Á¶°ñ¼¼Æ÷¸¦ 3ÀÏ ¹è¾çÇÑ °æ¿ì, ¿À½ºÅ׿ÀÄ®½Å »ý¼º¾çÀº 1.0 mMÀÇ Àú³óµµ¿¡¼­µµ °¨¼ÒÇÏ¿´´Ù. ´ÏÄÚƾÀÌ ¿À½ºÅ׿ÀÄ®½Å ´Ü¹éÁú »ý¼º°ú ¿À½ºÅ׿ÀÄ®½Å mRNA »ý¼º¿¡ ¹ÌÄ¡´Â ¿µÇâÀº 1ÀÏ°ú 3ÀÏ¿¡¼­ ´Ù¼Ò Â÷ÀÌ°¡ ÀÖ¾ú´Ù.
¿À½ºÅ׿ÀÇÁ·ÎÅ×Á¦¸°ÀÇ »ý¼º¾çÀº ¸ðµç ½ÇÇ豺¿¡¼­ ´ÏÄÚƾÀ» ÇÔÀ¯ÇÏÁö ¾ÊÀº ´ëÁ¶±ºº¸´Ù °¨¼ÒÇÏ¿´´Ù. ÇÏÁö¸¸ mRNA ¼öÄ¡´Â ´Ü¹éÁú »ý¼º°ú »ó¹ÝµÇ°Ô 7.5 mM (3ÀÏ), 5.0 mM (6ÀÏ) ÀÌ»óÀÇ °í³óµµ¿¡¼­ Áõ°¡ÇÏ¿´´Ù.

The purpose of this study was to evaluate the correlation between nicotine and the activity of bone forming cell. MG63 osteoblast-like cells were used for this study. Several factors were examined including the proliferation of cell, alkaline phosphatase activity, the formation of osteocalcin and osteoprotegerin, and the synthesis of its mRNA. MG63 osteoblast-like cells were incubated for 1, 2, 3 and 6 days with nicotine added to the culture medium in 1.0 ¥ìM, 1.0 mM, 2.5 mM, 5.0 mM, 7.5 mM, and 10.0 mM concentrations.
The proliferation of MG63 osteoblast-like cells was temporarily activated at the low nicotine concentrations. At high concentrations (£¾5.0 mM), however, it was suppressed. Alkaline phosphatase activity was suppressed in a dose-dependent manner as the concentration of nicotine increased.
Osteocalcin decreased in a dose-dependent manner at high nicotine concentrations of more than 7.5 mM and the same result was show when the osteoblasts were treated with low concentrations for longer than 3 days. There was a difference in the influence of nicotine on the synthesis of osteocalcin mRNA and formation of osteocalcin itself at 1 and 3 days.
Generally, osteoprotegrin notably declined in all experimental groups. However, the level of its mRNA increased at high nicotine concentrations of more than 7.5 mM after 3 days and more than 5.0 mM after 6 days.
